In order to successfully meet the aims and objectives of the project, it is essential to develop scientific methodologies,
study designs, interpret data, conduct the practical procedures systematically and in a planned manner.
My project incorporates the multi-disciplinary subject of pharmaceuticals, analytical chemistry, drug delivery, and
pharmacology. This area requires skills and extensive knowledge of the polymers, drug product formulations, drug
delivery and physical pharmacy. As well as, it includes characterizing the absorption and permeation of the API, their
diffusion into systematic circulation to elicit a pharmacological effect and how they are influenced by the oral enzymes
to form metabolites. There is a need to acquire the skills of in vitro or ex vivo methods for assessing buccal drug
permeation. This method involves the use of either excised porcine buccal tissue or buccal cell cultures (TR146 buccal
cell lines). The most significant question concerning the use of animal tissue in such a manner is the viability and
integrity of the dissected tissue. How well the dissected tissue is preserved is an important issue, which directly affects
the results and conclusion of the studies. Thus, it requires great attention and expertise for surgically removing the
underlying connective tissue from the oral cavity of pig and isolating the porcine buccal mucosal membrane with the
correct thickness. The general protocol for the use of animal tissues involves the sacrifice of the animal just before
experimentation and removal of the buccal mucosa with accompanying connective tissues, after which the buccal
epithelium is excised using surgical scissors. Here, excised animal buccal epithelium is mounted on perfusion/
diffusion chambers such as Franz cells, and permeation experiments are carried out, which compares the dimensions
and characteristics of available animal buccal tissues to that of humans. There is a requirement to possess the specific
set of skills in handling and operating several machines for the characterization of our model drug, both for quantitative
and qualitative analysis. As an example, Scanning electron microscopy (SEM) or Transmission electron microscopy
(TEM) for examining the particle surface morphology, FTIR for identifying the peaks corresponding to functional
groups present in the polymers, X-ray diffractometry (XRD) for investigating the absence or presence of crystallinity in
the polymers, circular dichroism (CD) for analyzing changes in protein secondary structural conformation, High
performance liquid chromatography (HPLC) for the separation, isolation and quantification of components of a
samples and so on. Then, the data after the experiments will be analyzed for statistical significance by using the
software like SPSS or GraphPad Prism®, thus, acquiring a proficiency in using these statistical tools is also a prerequisite
for computational data analysi
